Hi Sefika!

Calculation seems easiest/fastest.  You need to know what the weights of the copper are on each layer.  The board was built with foils having specifications of "ounces per square foot" of copper (as a US citizen I do feel guilty about these ridiculous units!).  Then plating will add a certain thickness.  A decent "CAM" program, such as Pentalogix' "ViewMaster Plus" has a utility to calculate the area of the copper on each layer from the photoplots.  Unless you have a very large ratio of vias to the area of the metal on each layer, you can ignore them.

You could probably do a similar calculation knowing all of the data on the particular laminate selected, but the variance will be higher.
