It will be very difficult to resolve variations in surface roughness of most metal plating using an optical microscope of any kind. You could try a SEM (scanning electron microscope), you'd be able to qualitatively 'measure' surface roughness with the higher magnification or resolution you can achieve with an SEM. Alternatively the most suitable instrument for your purposes would appear to be a Talysurf (http://www.taylor-hobson.com) profilometer, or similar, which will give a measurable micro profile or surface roughness measurement.
