You should work with your PCB house. Given your requirements and
needs, your PCB house should be able to deliver a working board on the
first or second pass. (Given that the board design can be adjusted to
meet your requirements!)

I have collected a good deal of data on the manufacture of
controlled impedance p.c.b's, but I am not sure where I should start.
Several people have told me I should start building test coupons
stepped out on a panel. Is this the right place to start? How would
I go about designing or obtaining a test coupon with a specific
characteristic impedance? I would appreciate any help I could get on
this matter. Thank you.
