Fabien,

If you are going to do destructive analysis I would recommend cutting the board close to a PTH and then use fine sand paper and grind into the PTH to expose the plating on the back wall of the PTH (i.e., make what looks like a castellation) and then do XRF measurements on the PTH barrel just like you would on the surface.

Vladimir is absolutely correct.
 
However, I will add that softer gold sandwiched between two hard nickel layers requires good sectioning skill and technique.
 
ENIG gold is 2-5 microinches in thickness (per IPC) so you will not only need good technique but good quality optics as well.
