Cadstar from Zuken does this. (www.cadstarworld.com)
Although it is in the way of net spacings, where you define the class of net & what the spacing for it should be.
I sometimes use it for crystal or mains tracks to keep everything else away.
However, it's a small matter to warrant the purchase of such a high end system.

Subject: [DC] Automatic Keepouts
Are there any pcb design programs that are capable of generating automatic
keepouts? ie: If I have a manually routed 80 mil trace which has a keepout
of 125 mils for U.L. spacing requirements, I'd like the software to place a
keepout around the entire trace automatically. This way, if I need to move
the trace at a later date, the keepout will move along with the trace. If
I change the size of the trace, the keepout would also 'recalculate and
regenerate' itself to retain the 125 mil spacing requirement.
