Since board edges are routed AFTER drilling,
it doesn't matter where you put the holes.
Unplated holes do not affect the router bit.
We have holes along the board edge on EVERY design

Plated holes are a different story, but many vendors
have no trouble creating "half-holes", which makes
the edge of the board look like a castellated component.
There are some extra process steps to keep the plated
barrel from ripping out, but it is done all the time.

Can anyone quote the spec for how close a nonplated thru hole can get to
the board edge.  I know the rule of thumb, but I need to see something in
writing.
